Arg5, derived from Archangium violaceum, encodes a heme-dependent tryptophan 4-hydroxylase that catalyzes the conversion of tryptophan to 4-hydroxytryptophan (4-HTP). In this reaction, the ferrous ion (Fe2+) within the heme cofactor activates molecular oxygen, thereby enabling regioselective hydroxylation. When heterologously expressed in Escherichia coli, the in vivo catalytic activity of Arg5 is markedly enhanced under conditions of increased aeration and is modulated by intracellular Fe2+ availability [339].
