The PsiM gene encodes an N-methyltransferase that catalyzes the methylation of norbaeocystin, yielding psilocybin as part of the native biosynthetic pathway. The enzyme displays strict substrate specificity toward the phosphorylated intermediate and is unable to catalyze the direct N-methylation of 4-hydroxytryptamine (4-HT). Substrate recognition and catalytic activity are mediated by specific interactions between conserved arginine residues (Arg75 and Arg281) and the phosphate moiety of the substrate [339].
