CipB is a surface-associated scaffold protein from Photorhabdus luminescens that binds defined domains of the outer-membrane adhesin CipA and serves as the primary docking element in a CipA–CipB scaffold system. Through this interaction, CipB anchors sequential enzymes and enables channel-like transfer of metabolic intermediates between consecutive catalytic steps. When used to recruit enzymes such as HisH/F or EcPurH, CipB scaffolding reduces accumulation of the by-product AICAR and enhances ATP synthesis. CipB efficiently incorporates exogenous proteins, forms physically independent complexes, and, when fused to fluorescent proteins such as mKate, displays homogeneous intracellular distribution and self-assembles into protein crystalline inclusions in E. coli [340].
