idi encodes isopentenyl diphosphate isomerase from Escherichia coli, which catalyzes the reversible interconversion of isopentenyl diphosphate (IPP) and dimethylallyl diphosphate (DMAPP) [354,347]. This reaction exhibits an equilibrium constant of approximately three, favoring conversion toward DMAPP [354]. Consequently, overexpression of idi has been widely employed in engineered strains to optimize the IPP/DMAPP ratio, thereby enhancing the supply of C5 isoprenoid precursors for downstream terpenoid biosynthesis, including geraniol and sesquiterpene production [354,347].
