pgl encodes 6-phosphogluconolactonase (PGL), which catalyzes the conversion of 6-phosphogluconolactone to 6-phosphogluconate in the oxidative branch of the pentose phosphate pathway [341, 345]. Overexpression of pgl increases flux through the pentose phosphate pathway and NADPH availability, thereby enhancing biomass accumulation and riboflavin production [345]. By contrast, pgl is absent in Escherichia coli BL21 (DE3), which restricts oxidative pentose phosphate pathway flux and shifts carbon flow toward the non-oxidative branch, increasing phosphoribosyl pyrophosphate (PRPP) availability and improving inosine monophosphate (IMP) biosynthesis [341].
