prs encodes phosphoribosyl pyrophosphate synthetase (Prs), which catalyzes the formation of 5-phospho-α-D-ribose-1-diphosphate (PRPP) from ribose-5-phosphate (R5P) and ATP, a key precursor in purine biosynthesis [340, 341]. Prs is feedback-inhibited by Mg-ADP, thereby coupling PRPP synthesis to the cellular energy status. In metabolic engineering, the feedback-insensitive prsD128A allele is employed to relieve this inhibition and increase PRPP availability [341].
