purR encodes a transcriptional regulator of purine biosynthesis that, in complex with hypoxanthine, represses the expression of multiple genes in the purine biosynthetic pathway in Escherichia coli [341, 345], including PRPP synthase (prs) [340]. Disruption of purR by pgl insertion may therefore enhance riboflavin production by derepressing purine precursor supply [345]. Deletion of purR increases intracellular PRPP levels, a key precursor for L-histidine and ATP biosynthesis [340].
